Summary: Genetic variations in the CACNA1C gene are strongly associated with psychiatric disorders such as schizophrenia and bipolar disorder. Studying the impact of mutations in Cacna1c on rat cognitive and synaptic functions revealed impairments in learning and synaptic plasticity, with dys-coordinated network oscillations and disrupted Ca2+ signaling pathways. Activation of the ERK pathway through small-molecule agonists rescued behavioral abnormalities in rats with Cacna1c mutations, suggesting targeted neurotrophin signaling pathways as a potential therapeutic approach for psychiatric disorders.
