Summary: Axon remodeling through sprouting and pruning is important for the refinement of developing neural circuits. Developing sensory axons deprived of neurotrophic support undergo apoptosis, with the tumor suppressor P53 playing a crucial role in activating the apoptotic pathway in axons. P53 appears to act in the cytoplasm to directly activate the mitochondrial apoptotic pathway in axons, highlighting a cytoplasmic role in the anterograde death of developing sensory axons.

Genetic loss-of-function experiments show that the small GTPase RhoA relays extracellular inhibitory signals to the cytoskeleton. Neuronal RhoA restricts axon regeneration by activating myosin II, while astrocytic RhoA limits injury-induced astrogliosis through YAP signaling.
