Reconnaissance of tumor immune microenvironment spatial heterogeneity in metastatic renal cell carcinoma and correlation with immunotherapy response

A. Hajiran et al.

Summary: A better understanding of the tumor immune microenvironment (TIME) in metastatic clear cell renal cell carcinoma (ccRCC) may inform precision treatment strategies. Through quantitative multiplexed immunofluorescence analysis, it was found that different immune marker densities in various regions (such as tumor core, tumor-stroma interface, and stroma) were associated with patient survival outcomes.

Spontaneous cell fusions as a mechanism of parasexual recombination in tumour cell populations

Daria Miroshnychenko et al.

Summary: The study demonstrates that spontaneous fusions between cancer cells can lead to partial recombination, increasing heterogeneity and phenotypic plasticity of tumor cells, accelerating their ability to adapt to new selective pressures.

Progressive immune dysfunction with advancing disease stage in renal cell carcinoma

David A. Braun et al.

Summary: The tumor immune microenvironment in clear cell renal cell carcinoma was investigated through single-cell RNA and T cell receptor sequencing, revealing an increase in terminally exhausted CD8(+) T cells and M2-like macrophages as the disease progresses. This immune dysfunction circuit is associated with a worse prognosis and identifies potential targetable immune inhibitory pathways in ccRCC.

Aberrant Epidermal Growth Factor Receptor RNA Splice Products Are Among the Most Frequent Somatic Alterations in Clear Cell Renal Cell Carcinoma and Are Associated with a Poor Response to Immunotherapy

Saif Zaman et al.

Summary: This study characterized a novel EGFR splicing alteration in renal cell carcinoma, which was specific to patients with advanced ccRCC and associated with poor response to immunotherapy. High expression of EGFR_pr20CTF correlated with lower survival rates and <16 months average survival in patients.

A comparison of immune receptor recombination databases sourced from tumour exome or RNAseq files: Verifications of immunological distinctions between primary and metastatic melanoma

Dhruv N. Patel et al.

Summary: It has been found that RNAseq and exome files prepared from tissue can be utilized for analyzing adaptive immune receptor recombination, adding value to datasets originally intended for gene expression or mutation studies. The two sets of IR recombination reads largely agree on several parameters, but using CDR3 chemical features for survival distinctions shows inconsistency. Recovery of TRB recombination reads from metastatic melanoma is associated with a better outcome, while the recovery of IR recombination reads from primary melanoma indicates a worse outcome.
