Summary: The study established a humanized mouse melanoma model and found high expression of chemokines in specific tumor regions, which is associated with resistance to PD-1 therapy. Further investigations showed reduced HLA-class I expression and CD8/Granz B+ T cell homeostasis in tumor areas where FOXP3 regulatory T cells and mast cells co-localize.

A pan-cancer single-cell transcriptional atlas of tumor infiltrating myeloid cells

Summary: This study conducted a pan-cancer analysis of single myeloid cells from 210 patients across 15 human cancer types, revealing distinct features of TIMs in different cancer types. For example, mast cells in nasopharyngeal cancer were associated with better prognosis, and pro-angiogenic TAMs showed diverse markers across different cancer types. The composition of TIMs appeared to be linked with certain features of somatic mutations and gene expressions.

The high-risk score was associated with shorter overall survival in BC patients, as well as cell proliferation and mast cell infiltration in Basal-like subtype BCs. The findings suggest that the prognostic signature based on CAF-secreted cytokines has implications for clinical outcome, tumor progression, and genetic alterations.
