Targeted reduction of cholesterol uptake in cholesterol-addicted lymphoma cells blocks turnover of oxidized lipids to cause ferroptosis

Summary: Recent research suggests that malignant cells dependent on cholesterol uptake from lipoproteins are susceptible to ferroptosis, a type of cell death triggered by accumulation of oxidized lipids in cell membranes. Treatment of these cells with HDL NPs activates a compensatory metabolic response, leading to increased de novo cholesterol synthesis and downregulation of GPX4 expression, resulting in ferroptosis-mediated cell death. In vivo studies in mouse lymphoma xenografts and patient samples further support the potential of targeting SCARB1 with HDL NPs for cholesterol uptake-addicted lymphoma cells.
