SETD8 potentiates constitutive ERK1/2 activation via epigenetically silencing DUSP10 expression in pancreatic cancer

Summary: In this study, it was found that SETD8 interacted with STAT3 on the DUSP10 promoter region to epigenetically silence DUSP10 expression, leading to enhanced ERK1/2 activation and unfavorable prognosis in pancreatic adenocarcinoma (PDAC). This study highlights the role of epigenetic mechanisms in modulating ERK1/2 activation and cell proliferation in PDAC.

Summary: Studies have shown that E2F1 plays a key role not only in regulating the cell cycle, but also in inducing lipid accumulation and elevated levels of lipogenic enzymes in ccRCC cells by upregulating SREBP1, promoting tumor proliferation and metastasis. Knockdown of E2F1 or suppression of SREBP1 can attenuate these effects in ccRCC cells.
