Summary: Pancreatic neuroendocrine tumor (pNET) is a type of pancreatic tumor with neuroendocrine differentiation. Early stage pNET can be treated with surgical resection for long-term survival, while pNET with metastasis has a poor prognosis. In this study, it was found that c-Myc overexpression is associated with lymph node metastasis in pNET xenograft mice. Combination therapy using an mTOR inhibitor and c-Myc inhibitor or VEGFC-neutralizing protein reduced lymph node metastasis in mice with c-Myc overexpression, indicating potential therapeutic targets for pNET.

circDCUN1D4 suppresses tumor metastasis and glycolysis in lung adenocarcinoma by stabilizing TXNIP expression

Summary: The aberrant expression of circRNAs plays a role in cancer progression through interactions with RNA-binding proteins (RBPs). A novel circRNA, circDCUN1D4, has been identified to be involved in the metastasis and glycolysis of lung adenocarcinoma (LUAD) by facilitating interactions between HuR and TXNIP proteins. It acts as a scaffold to enhance stability and transportation of TXNIP mRNA, ultimately suppressing metastasis and glycolysis of lung cancer cells in a TXNIP-dependent manner. The downregulated expression of circDCUN1D4 in lymph node metastatic tissues serves as an independent risk factor for the overall survival of LUAD patients.

SF3B1 mutation in pancreatic cancer contributes to aerobic glycolysis and tumor growth through a PP2A-c-Myc axis

Summary: The SF3B1(K700E) mutation promotes tumor growth in pancreatic ductal adenocarcinoma, while knockdown of SF3B1 has a significant inhibitory effect on cell proliferation in cell lines carrying this mutation. Ectopic expression of SF3B1(K700E) restores the inhibitory role induced by SF3B1 knockdown. The mutation also promotes tumor glycolysis through the aberrant splicing of PPP2R5A and activation of c-Myc. Pharmacological activation of PP2A with FTY-720 compromises the growth advantage induced by the SF3B1(K700E) mutation.
