Functional Genomics Identify a Regulatory Risk Variation rs4420550 in the 16p11.2 Schizophrenia-Associated Locus

Hong Chang et al.

Summary: This study comprehensively characterized the functional risk variation at 16p11.2 associated with schizophrenia by conducting multiple bioinformatics and molecular analyses. The results suggest that rs4420550 may play a key role in regulating the transcription and proliferation rates of genes at the 16p11.2 region.

Convergent lines of evidence support NOTCH4 as a schizophrenia risk gene

Yan Zhang et al.

Summary: The study provides convergent evidence supporting the association of NOTCH4 with schizophrenia and suggests a possible mechanism for its role in the pathogenesis of schizophrenia. Through meta-analysis, eQTL analysis, and integration of GWAS data, genetic variants in NOTCH4 were found to be significantly associated with schizophrenia risk, potentially through regulating NOTCH4 expression. Further experimental evidence showed that NOTCH4 dysregulation may affect neurodevelopment and contribute to schizophrenia risk.

Genome-wide association study followed by trans-ancestry meta-analysis identify 17 new risk loci for schizophrenia

Jiewei Liu et al.

Summary: This study identified two new schizophrenia risk loci in the Han Chinese population and 15 novel risk loci through a large-scale meta-analysis in East Asian and European populations. The study also found significant enrichment of schizophrenia heritability in certain genomic regions and tissue types. Additionally, the study highlights the importance of conducting genetic studies in diverse populations for a comprehensive understanding of schizophrenia genetics.
