The epigenetic basis of cellular heterogeneity

Benjamin Carter et al.

Summary: Single-cell sequencing methods have shown heterogeneity in gene expression levels among morphologically similar cells, with epigenomic information contributing to the regulation of gene expression.Recent studies suggest that variations in chromatin organization collectively define gene expression heterogeneity among otherwise similar cells.

Foxc1 establishes enhancer accessibility for craniofacial cartilage differentiation

Pengfei Xu et al.

Summary: The specification of cartilage relies on Sox9, but how transcription factors like Sox9 gain access to cartilage-specific regulatory regions during skeletal development was unknown. By analyzing chromatin accessibility in zebrafish chondrocytes, it was found that Foxc1 promotes chondrogenesis by establishing chromatin accessibility at cartilage-associated enhancers, with a decrease in accessibility leading to loss of dorsal facial cartilages. These findings highlight the dynamic establishment of cartilage-associated chromatin accessibility and the role of Foxc1 in promoting chondrogenesis in the face.

Chromatin Accessibility Identifies Regulatory Elements Predictive of Gene Expression and Disease Outcome in Multiple Myeloma

Benjamin G. Barwick et al.

Summary: Genetic and transcriptional characterization of multiple myeloma has identified subtypes with prognostic and therapeutic implications, while little is known about the myeloma epigenome. Research on the chromatin accessibility in myeloma cells from fresh and frozen samples revealed similarities in gene expression and highlighted the importance of enhancer activity in predicting oncogene expression and defining myeloma subtypes. This study demonstrates the feasibility of using biobanked specimens for retrospective studies on the myeloma epigenome and provides insights into unique enhancer landscapes in different myeloma subtypes.

Epigenetic mechanisms in breast cancer therapy and resistance

Liliana Garcia-Martinez et al.

Summary: The majority of breast cancers express the estrogen receptor and endocrine therapy has been the mainstay for hormone-responsive breast cancer treatment. Recent studies have shown the importance of the epigenome in determining hormone therapy response and how targeting epigenetic factors can help overcome clinical resistance. Understanding epigenetic mechanisms regulating ER+ breast cancer and endocrine therapy resistance is crucial for improving targeted therapies for this type of cancer.

Single cell transcriptional and chromatin accessibility profiling redefine cellular heterogeneity in the adult human kidney

Yoshiharu Muto et al.

Summary: The integration of single cell transcriptome and chromatin accessibility datasets provides deeper insights into cell heterogeneity, demonstrating the potential of single nucleus ATAC-seq in the identification of cell identity and functional heterogeneity. Differentially accessible chromatin regions in promoters associated with differentially expressed genes suggest the involvement of transcription factors like NF-kappa B in driving transitions between cell subpopulations within the kidney. This multi-omics approach improves the detection of unique cell states and redefines cellular heterogeneity in specific regions of the kidney.

Deficient H2A.Z deposition is associated with genesis of uterine leiomyoma

Davide G. Berta et al.

Summary: One in four women suffer from uterine leiomyomas (ULs) at some point in premenopausal life, which can cause excessive bleeding, pain, and infertility. Recent research identified mutations in genes encoding members of the SRCAP histone-loading complex as potential drivers of UL, indicating a potential mechanism of tumorigenesis involving epigenetic instability caused by deficient H2A.Z deposition.
