Phase II Trial of Costimulation Blockade With Abatacept for Prevention of Acute GVHD

Benjamin Watkins et al.

Summary: The study showed that adding abatacept to URD HCT was safe and effective in reducing AGVHD and improving SGFS. These results suggest that abatacept may have a significant positive impact on transplant outcomes related to AGVHD, particularly in HLA-mismatched HCT.

Impaired T- and NK-cell reconstitution after haploidentical HCT with posttransplant cyclophosphamide

Benedetta Rambaldi et al.

Summary: Administration of posttransplant cyclophosphamide has increased the number of patients undergoing HLA-haploidentical hematopoietic cell transplantation. Immune reconstitution in haplo-HCT patients showed delayed T-cell recovery skewed towards effector memory T cells and higher regulatory T cell ratios early after transplant. NK cell reconstitution also differed, with increased immature NK cells and enrichment of specific NK cell metadusters after haplo-HCT.

Metabolic programs define dysfunctional immune responses in severe COVID-19 patients

Elizabeth A. Thompson et al.

Summary: The study identifies a unique population of T cells expressing increased VDAC1 in severe and recovered COVID-19 patients, associated with mitochondrial dysfunction and apoptosis. Additionally, specific metabolic phenotypes of myeloid-derived suppressor cells in COVID-19 patients can distinguish between severe and mild disease. These findings offer insight into dysfunctional immune response in COVID-19 patients and potential for developing personalized therapeutic approaches.

Relationship of donor age and relationship to outcomes of haploidentical transplantation with posttransplant cyclophosphamide

Amy E. DeZern et al.

Summary: This study evaluated the impact of modifiable donor characteristics in haplo BMT and found that increasing donor age was associated with poorer outcomes, including worse survival, higher GVHD risk, and increased nonrelapse mortality. Younger adult-sized donors should be preferred when multiple haplo donors are available.
