The Multifaceted Role of Epoxide Hydrolases in Human Health and Disease

Jeremie Gautheron et al.

Summary: Epoxide hydrolases (EHs) are key enzymes involved in the detoxification of xenobiotics and biotransformation of endogenous epoxides, orchestrating crucial signaling pathways for cell homeostasis. The EH family comprises 5 proteins and 2 candidate members, with the best-known EHs being EPHX1 and EPHX2, which have wide expression patterns and multiple functions, leading to the development of specific inhibitors. Studies on pathogenicity of variants in EH genes are ongoing, highlighting the pleiotropic role of EHs in various disorders.

Summary: Chronic pain is a complex condition with significant physical, emotional, and financial impacts. Current treatments are insufficient due to high cost and safety issues. Inhibition of soluble epoxide hydrolase (sEH) as a therapeutic target could enhance the beneficial effects of natural epoxy-fatty acids (EpFAs) in pain management.

Summary: This report describes the development of an orally active analgesic that can address inflammation and neuropathic pain without causing addiction. The medication stabilizes EpFA to reduce pain and inflammation, with human trials showing no drug-related adverse events. Additionally, the fundamental research leading to the discovery of this medication and its mechanism of action is outlined.

Summary: The novel tolmetin derivative 5b exhibited high inhibitory activity against HL-60, HCT-15, and UO-31 tumor cell lines, as well as VEGFR-2. It induced apoptosis in HCT-15 cells, arrested the cell cycle in the G0/G1 phase, and displayed promising pharmacokinetic properties according to ADME studies.
