Summary: The market approval of Pitolisant has increased interest in clinical applications for novel multifunctional histamine H3 receptor antagonists. Various combinations of H3R pharmacophores with elements of other receptors, transporters, or enzymes have been synthesized. Recent studies suggest that some H3 receptor antagonists have affinity for sigma-1 receptor binding sites, which may impact their overall efficacy. The combination of histamine H3 and sigma-1 receptors could be a potential target for therapeutic developments in treating various diseases.

Structural modifications in the distal, regulatory region of histamine H3 receptor antagonists leading to the identification of a potent anti-obesity agent

Summary: A study on 4-pyridylpiperazine derivatives as histamine H-3 receptor ligands showed that the introduction of electron-withdrawing moieties into the distal aromatic ring significantly affected their affinity toward H3R. The 4-pyridylpiperazine moiety was found to be crucial for suitable interaction with the human histamine H-3 receptor, with even slight structural modifications leading to reduced affinity for H3R. Additionally, molecular modeling techniques were used to demonstrate the protein-ligand interactions responsible for the high affinity observed.
