Protein-protected metal nanoclusters as diagnostic and therapeutic platforms for biomedical applications

Gold Nanoclusters as Nanoantibiotic Auranofin Analogues

Summary: This study reports the design and synthesis of gold nanoclusters based on the structural motif of auranofin, which exhibit activity against both Gram-negative and Gram-positive bacteria, including multidrug-resistant pathogens. These AuNCs show enhanced antibacterial activity characterized by a greater uptake of gold, increased oxidative stress, moderate inhibition of thioredoxin reductase, and DNA damage. The uptake of AuNCs is not affected by the bacterial outer membrane barrier or extracellular protein binding, unlike Au-I complexes such as auranofin.
