Arrested Substrate Binding Resolves Catalytic Intermediates in Higher-Plant Water Oxidation

Georgia Zahariou et al.

Summary: The study reveals the importance of the S-3 state in the water-oxidizing Mn4CaO5 cluster of PSII, as it binds a substrate and precedes O-2 evolution. Methanol-treated PSII shows a previously uncharacterized high-spin species, coexisting with the known intermediate-spin conformation. These results provide insights into the structural heterogeneity of the S-3 state and offer constraints on the S-3 to S-4 transition and the mechanism of O-O bond formation.

Orientational Jahn-Teller Isomerism in the Dark-Stable State of Nature's Water Oxidase

Maria Drosou et al.

Summary: The study reveals that the tetramanganese-calcium cluster of the oxygen-evolving complex in photosystem II adopts orientational Jahn-Teller isomeric forms in the resting state, which explains previous spectroscopic observations and establishes the functional role of this isomerism in the catalytic process.

Proton exit pathways surrounding the oxygen evolving complex of photosystem II

Divya Kaur et al.

Summary: Photosystem II uses water as the primary electron source for photosynthesis, with water-filled channels playing a crucial role in bringing in and transporting water molecules. Analysis shows that the water chain in the broad channel is better connected, facilitating proton translocation. However, the specific roles of each pathway in the oxidation and reduction processes still need further investigation.

Summary: Arabidopsis thaliana, the first higher plant with a fully sequenced genome, lacks a high-resolution structure of Photosystem II; our study presents the first Cryo-EM high-resolution structure of Arabidopsis PSII supercomplex. The digitonin extracted PSII complexes elucidate key structural and stabilizing factors of Photosystem II.
