Amino Acid Sensing in Metabolic Homeostasis and Health

Xiaoming Hu et al.

Summary: The body's ability to sense and respond to changes in nutrient levels, particularly amino acids, is crucial for survival. Amino acids play dual roles as building blocks for proteins and peptides, as well as important bioactive molecules involved in signaling pathways and metabolic regulation. Imbalance in amino acid sensing signals may be linked to metabolic disorders, highlighting their potential as therapeutic targets.

Hepatic deletion of Mboat7 (LPIAT1) causes activation of SREBP-1c and fatty liver

Mingfeng Xia et al.

Summary: Recent identification of genetic variants that increase the risk of fatty liver disease and cirrhosis has led to the characterization of Mboat7 liver-specific KO mice in order to elucidate the link between these variants and fatty liver disease. The study revealed a relationship between fatty acid composition and regulation of hepatic fat synthesis, as well as delineated the mechanism by which mutations in MBOAT7 cause hepatic steatosis. Hepatic removal of both SREBP cleavage-activating protein (Scap) and Mboat7 normalized hepatic triglycerides relative to Scap-only hepatic KO, showing that increased SREBP-1c processing is required for Mboat7-induced steatosis.

The Power of Plasticity?Metabolic Regulation of Hepatic Stellate Cells

Parth Trivedi et al.

Summary: Hepatic stellate cells play a crucial role in regulating physiological and pathological responses in the liver, particularly in chronic liver injury. They drive hepatic fibrosis and are involved in inflammation and cancer by adapting metabolic pathways to meet increased energy demands.

Insulin action, type 2 diabetes, and branched-chain amino acids: A two-way street

Phillip J. White et al.

Summary: The association of obesity and insulin resistance with increased levels of branched-chain and aromatic amino acids and decreased glycine levels is well-established. Human metabolomics and genetic studies have further confirmed these observations, with preclinical studies identifying mechanisms involved in amino acid homeostasis disruption and its connection to dysregulated glucose and lipid metabolism, particularly in the development of insulin resistance, type 2 diabetes, and cardiovascular diseases.

SREBP-1c impairs ULK1 sulfhydration-mediated autophagic flux to promote hepatic steatosis in high-fat-diet-fed mice

Thuy T. P. Nguyen et al.

Summary: The study demonstrates that high-fat-diet induced SREBP-1c impairs autophagic lipid catabolism by altering H2S signaling, leading to hepatic lipid accumulation. This mechanism involves the reduction of CSE via miR-216a, which decreases hepatic H2S levels and sulfhydration-dependent activation of ULK1, contributing to the pathogenesis of NAFLD. The findings highlight a 2-fold mechanism for SREBP-1c-driven hepatic lipid accumulation through regulation of lipid biosynthesis and degradation.

SREBP1-induced fatty acid synthesis depletes macrophages antioxidant defences to promote their alternative activation

Guillaume Bidault et al.

Summary: Macrophages have a spectrum of activation states and play various roles in physiological processes. The study found that the anabolic transcription factor SREBP1 is essential for macrophage alternative activation by triggering the de novo lipogenesis program. Reactive oxygen species also play a critical role in signaling for macrophage alternative activation.
