Low modification of PETase enhances its activity toward degrading PET: Effect of conjugate monomer property

Fungal Hydrophobin RolA Enhanced PETase Hydrolysis of Polyethylene Terephthalate

Summary: Polyethylene terephthalate (PET) is a well-known polyester used as packaging material, and PETase has emerged as a potential biocatalyst for degrading PET. The presence of hydrophobin RolA significantly increased the rate of PETase hydrolysis, while enhancing the hydrolysis of PET bottles. This study suggests that RolA's wetting effect on the PET surface may convert PET to become hydrophilic, making it easier for PETase to contact and attack the surface.

Complicated effects of a zwitterionic polymer containing dimethyl chains on the structures, activities and stabilities of different enzymes

Summary: Zwitterionic polymer containing dimethyl chains (pID) has complex effects on the activity, stability, and structure of three different enzymes. Activity assays show that pID decreases LYS activity but increases CRL and CAT activities, while spectroscopic analysis reveals a static quenching mechanism of enzyme fluorescence by pID binding.

Polymer Modification of Lipases, Substrate Interactions, and Potential Inhibition

Summary: In this study, the industrially important enzyme, Candida antarctica lipase B (CalB), was modified with various functional polymers using a grafting approach. The impact of polymer chain length on CalB activity was investigated, revealing significant enhancements for acrylamide (Am) and N,N-dimethyl acrylamide (DMAm) conjugates in both aqueous and aqueous-organic media mixtures. Furthermore, the study demonstrated a gradual increase in activity for N,N-dimethylaminopropyl-acrylamide (DMAPA) conjugates with higher organic solvent content, contrary to diminished activity observed for hydrophobic polymer-protein conjugates. Overall, this research provides insights into potential advancements in lipase production, leading to new and improved lipase bioconjugates with enhanced activity and stability.

Highly Crystalline Post-Consumer PET Waste Hydrolysis by Surface Displayed PETase Using a Bacterial Whole-Cell Biocatalyst
