Summary: Programming non-canonical organisms for high-value chemical production is increasingly attractive. Optimizing heme metabolism in Shewanella oneidensis MR-1 revealed significant changes in amino acid profiles and increased production of the valuable compound 5-aminolevulinic acid (ALA) through CRISPR interference and modular design strategies. Integration of gene clusters under dual T7 promoters led to a remarkable 145-fold improvement in ALA production, demonstrating successful rewiring of metabolic flux in MR-1 for the first time.
