CRISPR-Act3.0 for highly efficient multiplexed gene activation in plants

Summary: A highly robust CRISPRa system, CRISPR-Act3.0, has been developed for efficient gene activation in rice, Arabidopsis, and tomato, resulting in fourfold to sixfold higher activation than current state-of-the-art CRISPRa systems. The system enables robust activation of up to seven genes in rice and simultaneous modification of multiple traits in Arabidopsis, stably transmitted to the T3 generations. Targeting rules for effective transcriptional activation have been elucidated, and further improvements and new systems have been developed based on the CRISPR-Act3.0 platform, providing plant researchers with a powerful toolbox for gene activation in foundational and translational research.
