Summary: The complement system, as an unexpected, cell-autonomous orchestrator of normal cell physiology, controls key pathways of metabolism during immune cell homeostasis and effector function. Little is known about the exact structure and localization of intracellular complement components, but they do interact closely with mitochondria, impacting metabolism significantly.

Complement C5 activation promotes type 2 diabetic kidney disease via activating STAT3 pathway and disrupting the gut-kidney axis

Summary: The study found that in diabetic kidney disease (DKD), high activation of C5 promotes disease progression by activating STAT3 and impairing the gut-kidney axis. The C5a receptor antagonist (C5aRA) can improve renal function damage, ECM deposition, macrophage infiltration, and proinflammatory factor expression in DKD mice, and partially restore gut microbiota diversity and SCFA levels.

Processed foods drive intestinal barrier permeability and microvascular diseases

Summary: The intake of processed foods has been associated with an increased risk of chronic kidney disease and diabetes. In rodent models, a processed diet was found to increase intestinal barrier permeability and the risk of CKD, while inhibiting the advanced glycation pathway reversed kidney injury. A high resistant starch fiber diet was shown to maintain gut barrier integrity and reduce severity of kidney injury in a mouse model of diabetes by suppressing complement activation. These findings provide insights into how processed foods contribute to chronic inflammation and disease.
