Reduced proinsecticide activation by cytochrome P450 confers coumaphos resistance in the major bee parasite Varroa destructor

Spyridon Vlogiannitis et al.

Summary: Varroa destructor mites from the island Andros exhibit high levels of resistance to coumaphos, with reduced activation of the proinsecticide by a cytochrome P450 enzyme being the main resistance mechanism. Transcriptome analysis revealed overexpression of CYP4DP24 and underexpression of CYP3012A6 and CYP4EP4 in resistant mites. RNA interference of CYP4EP4 in susceptible mites prevented coumaphos activation and decreased toxicity, suggesting a potential solution for overcoming resistance in Varroa destructor populations.

Heterologous expression of insect P450 enzymes that metabolize xenobiotics

Ralf Nauen et al.

Summary: Insect cytochrome P450 monooxygenases play a crucial role in oxidative transformation of substrates, including insecticides, detoxification of plant secondary metabolites, and determining insecticide selectivity in beneficial arthropods. Recent advancements in studying the functional relevance of P450s for xenobiotic metabolism through recombinant expression have shown partial success in improving functional expression by borrowing strategies from mammalian P450s.

Short term transcriptional responses of P450s to phytochemicals in insects and mites

Marilou Vandenhole et al.

Summary: Cytochrome P450 monooxygenases play a key role in detoxifying phytochemicals in arthropod herbivores. Recent research has revealed the gene expression plasticity of P450s in responding to phytochemicals, and whole genome transcriptomic analysis can help identify responsive P450 genes without bias.

Summary: The metabolic resistance to pyrethroids in the cotton bollworm has evolved through a unique chimeric enzyme, CYP337B3, which efficiently hydroxylates fenvalerate at the 4'-carbon position. Site-directed mutagenesis and docking models revealed the specific amino acids and structural features that enable CYP337B3 to exhibit this unique enzymatic function.
