Allosteric Modulation of GPCRs of Class A by Cholesterol

Jan Jakubik et al.

Summary: G-protein coupled receptors (GPCRs) are membrane proteins that transmit extracellular signals to cells, and membrane cholesterol plays a significant role in regulating their function through direct binding and membrane property changes. Cholesterol affects ligand binding and activation of GPCRs, as well as altering their oligomerization state. Manipulating membrane cholesterol levels or modulating cholesterol binding sites on GPCRs may have potential for the development of new therapies.

Direct Observation of Cholesterol Dimers and Tetramers in Lipid Bilayers

Matthew R. Elkins et al.

Summary: Using solid-state nuclear magnetic resonance (NMR) spectroscopy and molecular dynamics simulations, the study identified the oligomeric structure of cholesterol in phospholipid bilayers, confirming dimers as the basic structural unit. This has implications for understanding cholesterol interactions with membrane proteins and its role in diseases.

Pore-forming transmembrane domains control ion selectivity and selectivity filter conformation in the KirBac1.1 potassium channel

Marcos Matamoros et al.

Summary: The study showed that mutations can change the selectivity of potassium channels, and under high Na+/low K+ conditions, the SF can transition to dilated states. I131 and other structural elements play a crucial role in controlling ion selectivity, suggesting that their physical interaction with the SF contributes to the relative stability of the K+-induced SF configuration.

Cholesterol Is a Dose-Dependent Positive Allosteric Modulator of CCR3 Ligand Affinity and G Protein Coupling

Evan van Aalst et al.

Summary: Cholesterol serves as an allosteric modulator of G protein-coupled receptor function, influencing receptor-ligand interactions and signal transduction. CCR3, a chemokine receptor, plays a vital role in cell trafficking and is implicated in cancer metastasis and inflammatory conditions. However, there is a lack of experimental evidence linking the functional states of CCR3 to the molecular mechanisms of cholesterol-receptor cooperativity.
