Super-resolution imaging reveals α-synuclein seeded aggregation in SH-SY5Y cells

Jason C. Sang et al.

Summary: Researchers found that cells respond to disruption of protein homeostasis by alpha-synuclein seeds primarily by secreting nanoscopic aggregates. This mechanism may represent an important protective response by cells to protein aggregation.

Site-Specifically-Labeled Antibodies for Super-Resolution Microscopy Reveal In Situ Linkage Errors

Susanna M. Frueh et al.

Summary: In this study, a simple two-step protocol was developed to site-specifically attach reporters to IgG antibodies, and their performance in super-resolution microscopy techniques was evaluated. The results showed that errors from primary and secondary antibodies did not add up, and simulations revealed the main factors contributing to the errors.

The expanding amyloid family: Structure, stability, function, and pathogenesis

Michael R. Sawaya et al.

Summary: The hidden world of amyloid biology has revealed over 80 amyloid protein fibrils, showing their significance in diseases and biological functions. These proteins differ from globular proteins by stacking into unbranched fibrils, with individual protein sequences adopting different conformations. In functional amyloids, the conformation determines biological function, showcasing the versatility of the amyloid fibril scaffold.

Versatile naphthalimide tetrazines for fluorogenic bioorthogonal labelling

Marcus E. Graziotto et al.

Summary: This study presents a series of naphthalimide tetrazines for bioorthogonal fluorogenic labeling, which can be used for imaging and successfully localized in lysosomes and mitochondria with targeted variants. The naphthalimide tetrazine system was also utilized for no-wash imaging of insulin amyloid fibrils, offering a new method to monitor their growth kinetics. The synthetic approach used in this research provides a novel scaffold for a variety of bioorthogonal tetrazine-based imaging agents.
