Use of CRISPR/Cas9-mediated disruption of CNS cell type genes to profile transduction of AAV by neonatal intracerebroventricular delivery in mice

Tess Torregrosa et al.

Summary: The study combined CRISPR/Cas9 technology with AAV delivery of specific gene disruption in the CNS of mice, allowing for an accurate assessment of AAV tropism in neurons, astrocytes, and oligodendrocytes. AAV-PHP.B and AAV-PHP.eB showed significantly improved disruption of the NeuN gene compared to AAV9 in various brain regions and spinal cord, while all three AAV variants displayed modest disruption of the GFAP and MOG genes. The results suggest that AAV-PHP.B variants exhibit enhanced neuronal transduction in the CNS, regardless of their ability to cross the blood-brain barrier.

Adeno-associated virus capsid assembly is divergent and stochastic

Tobias P. Woerner et al.

Summary: Recent research on Adeno-associated viruses (AAVs) using high-resolution native mass spectrometry has revealed that the capsids of different AAV serotypes assemble stochastically, leading to a highly heterogeneous population of capsids with varying compositions. Even the most abundant VP stoichiometry represents only a small percentage of the total AAV population. The systematic scoring of simulations against experimental native MS data offers a sensitive new method to characterize these therapeutically important heterogeneous capsids.

Comprehensive characterization and quantification of adeno associated vectors by size exclusion chromatography and multi angle light scattering

Nicole L. McIntosh et al.

Summary: Size exclusion chromatography coupled to multiangle light scattering is a straightforward method for comprehensive characterization of AAV capsids, providing detailed information with high accuracy and precision. By using new equations and intrinsic properties, multiple physical attributes of AAV can be quantified in a short time, making this method a powerful tool for future gene therapy programs.

Optimized Reversed-Phase Liquid Chromatography/Mass Spectrometry Methods for Intact Protein Analysis and Peptide Mapping of Adeno-Associated Virus Proteins

Ximo Zhang et al.

Summary: The study developed reversed-phase liquid chromatography/mass spectrometry-based methods for characterizing AAV capsid proteins at intact protein and peptide levels with reduced sample consumption. Additionally, a 1-hour enzymatic digestion method was also developed using 1.25 µg of AAV VPs, providing over 98% protein sequence coverage and reproducible relative quantification of various PTMs of the VPs. These methods enable efficient and sensitive analyses of AAV capsid proteins, enhancing understanding and guiding the development and manufacturing of AAV-related therapeutics.

Multi-dimensional LC-MS: the next generation characterization of antibody-based therapeutics by unified online bottom-up, middle-up and intact approaches

Julien Camperi et al.

Summary: The accelerated development of new therapeutics in a competitive landscape necessitates high throughput analytical platforms. Emerging formats of biotherapeutics have increased the complexity, emphasizing the need to enhance conventional liquid chromatography resolution. Liquid chromatography-mass spectrometry technologies, such as multi-attribute methods, show potential in complementing traditional analytical toolbox for monoclonal antibody characterization. Multi-dimensional LC-MS provides a promising approach to address the challenges faced with novel antibody formats by combining sample preparation and multi-level analysis in a single chromatographic setup.
