Integrated Strategy Drives Direct Infusion-Tandem Mass Spectrometry as an Eligible Tool for Shotgun Pseudo-Targeted Metabolomics of Medicinal Plants

Ke Zhang et al.

Summary: In this study, direct infusion (DI) was integrated into the concept of pseudo-targeted metabolomics (PTM) to develop a new strategy enabling shotgun PTM. By applying versatile MS/MSALL and online ER-MS technologies, quantitative data was obtained through SRM and structural annotation confidence was strengthened using breakdown graphs. Through in-depth investigation of Cistanche species, confirmatory chemical markers for species discrimination were identified, demonstrating the utility of the new strategy for shotgun PTM beyond Cistanche plants.

DaDIA: Hybridizing Data-Dependent and Data-Independent Acquisition Modes for Generating High-Quality Metabolomic Data

Jian Guo et al.

Summary: A novel metabolomic data acquisition workflow, named data-dependent-assisted data-independent acquisition (DaDIA), that combines DDA and DIA analyses was proposed to achieve better data quality in LC-MS-based metabolomics. Computational program DaDIA.R was developed to process heterogeneous DDA and DIA data for streamlined metabolite annotation. The DaDIA workflow demonstrated significantly improved data quality compared to conventional DDA or DIA metabolomics, with increased number of detected features and annotated metabolites.
