Untargeted UHPLC-ESI-QTOF-MS/MS analysis with targeted feature extraction at precursor and fragment level for profiling of the platelet lipidome with ex vivo thrombin-activation

Malgorzata Cebo et al.

Summary: The study analyzed the lipidome of platelets using advanced technology, revealing the distribution and changes of lipids in platelets in resting and activated states. The results showed that various lipid species undergo significant alterations during platelet activation, with some lipids serving as biomarkers of activation.

Isomer Selective Comprehensive Lipidomics Analysis of Phosphoinositides in Biological Samples by Liquid Chromatography with Data Independent Acquisition Tandem Mass Spectrometry

Peng Li et al.

Summary: Phosphoinositides (PIPx) are crucial in cellular functions like growth, proliferation, differentiation, and are regulated by fatty acyl chains, degree of phosphorylation, and phosphorylation positions. However, there is a lack of comprehensive analytical method for distinguishing all regioisomeric forms with different acyl side chains and degrees of phosphorylation. This study presents a powerful method for analyzing PIPx profiles in biological samples using a chiral polysaccharide stationary phase and high-resolution mass spectrometry.

Current Challenges and Recent Developments in Mass Spectrometry-Based Metabolomics

Stephanie L. Collins et al.

Summary: High-resolution mass spectrometry has revolutionized the study of metabolism in living systems, allowing for the measurement of numerous metabolites in a single experiment. Despite improvements in mass detector sensitivity, challenges remain in metabolite identification, differentiation from environmental contaminants, chromatographic separation, and incomplete databases. Liquid chromatography-mass spectrometry-based methods are popular for their sensitive detection of small molecules, and advancements in instrumentation, experimental techniques, and computational tools have been developed to address these challenges.
