Summary: Vascular smooth muscle cells play major roles in pathological vascular remodelling, with clonal lineage tracing revealing that their contribution to disease results from hyperproliferation of a few pre-existing medial cells that can adopt diverse phenotypes. Studies have shown that VSMC-derived cells in vascular lesions have both beneficial and detrimental effects on disease severity. Computational analyses suggest that the pathway from healthy arteries to phenotypically distinct lesional cells consists of multiple, potentially regulatable, steps.

Summary: This review discusses the evolutionary role of these molecules in mechanosensing, limitations in identifying mechanosensitive molecules, and introduces a paradigm shifting molecular model for a VSMC mechanosensor. Loss of pressure-induced vasoconstriction can increase susceptibility to renal and cerebral vascular injury, with favored paradigms including transient receptor potential channels, integrins, and Degenerin channels as transducers.

Review of the Techniques Used for Investigating the Role Elastin and Collagen Play in Arterial Wall Mechanics

Summary: The arterial wall is composed of various components such as collagen and elastin fibers, proteoglycans, Vascular Smooth Muscle Cells (VSMCs) and ground matrix, which play different roles in determining the mechanical properties of the vascular tissue. Experimental methods including microscopy imaging and mechanical testing have been used to study the mechanical role of these structural proteins, but combining these methods provides a more complete understanding of the arterial micromechanics. Advances in imaging techniques allow for dynamic imaging of samples under a pseudo-physiological condition, overcoming the limitations of using either method separately. This review aims to describe the techniques available for studying arterial wall micromechanics and to identify gaps in current research on arterial mechanics.
