Evidence for a Geroscience Approach to Late Life Depression: Bioenergetics and the Frail-Depressed

Frailty and Depression in Late Life: A High-Risk Comorbidity With Distinctive Clinical Presentation and Poor Antidepressant Response

Summary: This study investigated the long-term relationship between physical frailty and antidepressant medication response in older adults with depressive illness. The results showed that antidepressant medication is less effective for treating major depressive disorder in frail older adults, and even when a response is achieved, it does not improve their frailty. These findings highlight the importance of assessing frailty in depressed older adults and the need for novel therapeutic strategies to address comorbid frailty and depression.

Increased Levels of Circulating Cell-Free mtDNA in the Plasma of Subjects With Late-Life Depression and Frailty: A Preliminary Study

Summary: This study evaluated the levels of circulating cell-free mitochondrial DNA (ccf-mtDNA), a marker of cellular stress and damage, in older adults with late-life depression (LLD) and frailty. The coexistence of LLD and frailty was associated with higher levels of ccf-mtDNA, suggesting a common biological mechanism of cellular stress and mitochondrial dysfunction. This finding offers potential opportunities for future interventions guided by geroscience.
