The emerging role and significance of circular RNAs in viral infections and antiviral immune responses: possible implication as theranostic agents

Faryal Mehwish Awan et al.

Summary: circRNAs, formed by back-splicing of pre-mRNAs, show high stability and resistance to degradation, which may have potential therapeutic applications. Despite their diverse biological roles, the circular structure and sequence overlap with linear mRNA counterparts make circRNAs challenging to study in depth, especially their involvement in viral infections and immune responses. Understanding circRNA-mediated viral protein regulations could enhance knowledge of these novel molecules and their implications for antiviral immunity and therapy.

A short ORF-encoded transcriptional regulator

Minseob Koh et al.

Summary: Recent technological advances have enabled the identification of previously unidentified short ORF-encoded peptides (SEPs) in mammalian genomes. By incorporating the photo-crosslinking noncanonical amino acid AbK into SEP transgenes, researchers have demonstrated a method to easily identify SEP cellular interaction partners and discovered a short ORF-encoded histone binding protein (SEHBP) that interacts with chromatin-associated proteins. This work provides a straightforward method to define the physiological roles of SEPs and highlights the utility of identifying SEHBP as a short ORF-encoded transcription factor.

lncRNA MIR155HG Accelerates the Progression of Sepsis via Upregulating MEF2A by Sponging miR-194-5p

Chao Zhang et al.

Summary: This study demonstrates that MIR155HG promotes sepsis progression by modulating the miR-194-5p/MEF2A axis. Knockdown of MIR155HG improves cell viability, reduces apoptosis and inflammation, which can be counteracted by upregulating miR-194-5p. Additionally, overexpression of MEF2A reverses the inhibitory effect on sepsis progression induced by MIR155HG deletion.

Summary: The study identified a high-throughput screening method to systematically discover RNA sequences that can direct circular RNA (circRNA) translation. Complementarity of 18S rRNA and structured RNA elements play important roles in driving circRNA translation. CircRNA-encoded proteins have tissue-specific distribution and play a role in antigen presentation.

Nuclear compartmentalization as a mechanism of quantitative control of gene expression

Prashant Bhat et al.

Summary: Gene regulation requires coordination of hundreds of regulatory factors at precise genomic and RNA targets. An emerging explanation suggests that DNA, RNA, and proteins organize into 3D compartments in the nucleus to concentrate functionally related molecules, leading to the formation of condensates that partition the nucleus. This compartmentalization can result in non-stoichiometric molecular interactions and behaviors.
