Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Adaptive and Constitutive Activations of Malic Enzymes Confer Liver Cancer Multilayered Protection Against Reactive Oxygen Species

Derek Lee et al.

Summary: This study reveals the distinct regulatory mechanisms of malic enzymes ME1 and ME3 in hepatocellular carcinoma (HCC), with ME1 being controlled by the NRF2-mediated oxidative stress response pathway and ME3 being constitutively induced by superenhancers. Disrupting these regulatory pathways can slow down HCC progression, and targeting both mechanisms may present a potential therapeutic approach for HCC treatment.

FOXP4-AS1 is a favorable prognostic-related enhancer RNA in ovarian cancer

Tian Hua et al.

Summary: This study identified prognostic-related eRNAs in ovarian cancer using integrated data analysis. Among 71 eRNA candidates, 23 were found to be correlated with their potential target genes, with FOXP4-AS1 showing the highest positive correlation with FOXP4. The results were further validated in an independent ovarian cancer cohort, suggesting FOXP4-AS1 as a favorable independent prognostic biomarker candidate in ovarian cancer.

Advances in immunotherapy for hepatocellular carcinoma

Bruno Sangro et al.

Summary: Immunotherapeutic interventions have shown effectiveness in the treatment of hepatocellular carcinoma. Checkpoint inhibitors have demonstrated strong anti-tumor activity, but other immune strategies have not yet shown consistent clinical activity. Advancing treatment to earlier stages, discovering predictive biomarkers, and exploring effective combinatorial approaches are key challenges in HCC immunotherapy.

LINC02257, an Enhancer RNA of Prognostic Value in Colon Adenocarcinoma, Correlates With Multi-Omics Immunotherapy-Related Analysis in 33 Cancers

Junbo Xiao et al.

Summary: Evidence supports the significant involvement of long non-coding RNAs (lncRNAs) in human cancer development, with attention now turning to the role of Enhancer RNAs (eRNAs) in carcinogenesis. In colon adenocarcinoma, LINC02257 emerges as a key eRNA with strong associations to unfavorable prognosis, particularly through its impact on extracellular matrix organization via the PI3K-Akt signaling pathway. Additionally, LINC02257 shows potential as an independent prognostic biomarker and an immunotherapy-related eRNA in various cancers.
