Revealing the Dynamic Mechanism by Which Transferrin Promotes the Cellular Uptake of HAIYPRH Peptide-Conjugated Nanostructures by Force Tracing

Summary: The peptide HAIYPRH (T7) is commonly used as a ligand in tumor-targeted nanodrug delivery systems. This study used atomic force microscopy to track the dynamic process of a T7-conjugated gold nanoparticle (AuNP-T7) entering cells, revealing that transferrin can both decrease endocytosis force and increase endocytosis speed depending on the cell type. This research provides important insights for the redesign and development of T7-conjugated nanodrug carriers in targeted nanodrug delivery systems.

Summary: Functionalization of nanoparticles with triphenylphosphonium (TPP) targeting moiety was explored in this study, revealing the influence of protein corona from fetal bovine serum and human plasma on targeting ability and cytotoxicity. The findings demonstrated that the protein corona could mitigate cytotoxicity of nanoparticle functionalization, but high protein density might reduce the targeting specificity at the subcellular level.
