Combinatorial Photothermal 3D-Printing Scaffold and Checkpoint Blockade Inhibits Growth/Metastasis of Breast Cancer to Bone and Accelerates Osteogenesis

Chao He et al.

Summary: A BG@NbSiR scaffold has been developed to effectively treat bone metastasis of breast cancer by activating immune response, suppressing metastases, preventing tumor relapses, and promoting osteogenesis. Single-cell sequencing has been used to identify critical factors responding to this scaffold-based therapy, providing insights into underlying therapeutic mechanisms.

Molybdenum derived from nanomaterials incorporates into molybdenum enzymes and affects their activities in vivo

Mingjing Cao et al.

Summary: Understanding the in vivo biotransformation of nanomaterials, such as molybdenum derived from MoS2, can shed light on their long-term effects on liver metabolism. Through a combination of in vivo experiments and molecular dynamics simulations, the study reveals that the biodistribution of molybdenum is mediated by protein coronas, mainly with apolipoprotein E, leading to increased enzyme activities in the liver. The findings suggest that nanomaterials consisting of essential trace elements may be converted into active biological molecules in organisms.

Covalent Bisfunctionalization of Two-Dimensional Molybdenum Disulfide

Xin Chen et al.

Summary: This study demonstrates a novel approach to constructing bisfunctionalized MoS2 hybrid structures by successively reacting activated MoS2 with alkyl iodide and aryl diazonium salts, showing higher reactivity compared to adducts formed through reactions with diazonium salts. The systematic study on formation and reactivity of covalently functionalized MoS2 hybrids provides practical guidance for tailoring the properties of 2D MoS2 for various potential applications.

Combination of optimized tissue engineering bone implantation with heel-strike like mechanical loading to repair segmental bone defect in New Zealand rabbits

Cong Zhu et al.

Summary: This study investigated the effects of combining optimized tissue engineering bone (TEB) implantation with heel-strike like mechanical loading to repair segmental bone defects in New Zealand rabbits. It was found that BMMSC-CBCs showed better proliferation and osteogenic differentiation abilities as seed cells for optimized TEB construction. The combination of optimized TEB implantation and heel-strike like mechanical loading had a synergistic effect on bone defect healing, with enhanced expression of TGFβ1 and BMP2 playing a key role in the process.

TRPM7 kinase-mediated immunomodulation in macrophage plays a central role in magnesium ion-induced bone regeneration

Wei Qiao et al.

Summary: This study reveals a biphasic mode of action of magnesium ion (Mg2+) in bone repair. During the early inflammation phase, Mg2+ can contribute to pro-osteogenic effects by promoting an immune microenvironment for bone formation; however, in the later remodeling phase, Mg2+ has negative effects on bone maturation and repair.

Yan Lu et al.

Summary: The study demonstrated that functionalizing collagen scaffolds with porcine decellularized extracellular matrix can promote neocartilage regeneration. Results showed that the solution form of ECM performed better in cell recruitment, proliferation, and chondrogenesis, quickly providing a favorable chondrogenic microenvironment for endogenous BMSCs.

Progenitor cell-derived exosomes endowed with VEGF plasmids enhance osteogenic induction and vascular remodeling in large segmental bone defects

Yao Zha et al.

Summary: In this study, a cell-free tissue engineering system using functional exosomes was developed to induce osteogenic differentiation of mesenchymal stem cells and release the VEGF gene for vascular remodeling. The engineered exosomes effectively combined with 3D-printed porous bone scaffolds and induced vascularized bone regeneration in vivo. This work highlights the potential of exosomes as therapeutic nanoparticles for acellular tissue engineering.
