Intranasal delivery of an antisense oligonucleotide to the RNA-binding protein HuR relieves nerve injury-induced neuropathic pain

Vittoria Borgonetti et al.

Summary: Neuropathic pain is a condition that is often not effectively treated, and there is a need for the development of new treatments. This study investigated the role of the RNA binding protein HuR in promoting neuropathic pain and trauma-induced microglia activation, using antisense oligonucleotide knockdown of HuR gene expression. The study found that intranasal administration of the antisense oligonucleotide effectively reduced spinal HuR protein levels and alleviated pain hypersensitivity, with similar efficacy to intrathecal delivery.

Cold-Shock Domains-Abundance, Structure, Properties, and Nucleic-Acid Binding

Udo Heinemann et al.

Summary: The cold-shock domain is a conserved protein domain with a simple architecture but complex biological functions, found from bacteria to humans, that can bind both DNA and RNA through non-specific molecular interactions. The structural properties of cold-shock domains allow them to function in transcription regulation, RNA splicing, translation, stability, and sequestration, primarily through promiscuous DNA and RNA binding.

HuR up-regulates cell surface PD-L1 via stabilizing CMTM6 transcript in cancer

Yanbin Liu et al.

Summary: The study revealed a positive correlation between CMTM6 and HuR expression in most types of cancer, where HuR stabilizes CMTM6 mRNA through direct association with AREs and upregulates CMTM6 expression, which can be inhibited by MS-444. In HuR-overexpressing cancer cells, abundant cell surface PD-L1 inhibits immune activation of T cells, but treatment with MS-444 relieves immune suppression and stimulates immune responses. Ectopic HuR accelerates allograft tumor progression in vivo, which can be greatly reduced by simultaneous administration of MS-444, suggesting the potential of combining HuR inhibitor with PD-1/PD-L1 antibodies for cancer immunotherapy.

Downregulation of hsa_circ_0074854 Suppresses the Migration and Invasion in Hepatocellular Carcinoma via Interacting with HuR and via Suppressing Exosomes-Mediated Macrophage M2 Polarization

Yong Wang et al.

Summary: The study revealed upregulation of hsa_circ_0074854 in HCC, with its knockdown inhibiting tumor growth. Mechanistically, hsa_circ_0074854 knockdown reduced HCC cell migration and invasion by interacting with HuR. Moreover, hsa_circ_0074854 was transferred from HCC cells to macrophages via exosomes, suppressing macrophage M2 polarization and subsequently inhibiting HCC cell migration and invasion.

SRSF1 inhibits autophagy through regulating Bcl-x splicing and interacting with PIK3C3 in lung cancer

Yuesheng Lv et al.

Summary: SRSF1 is identified as an autophagy suppressor that inhibits autophagosome formation by promoting the splicing of Bcl-x and disrupting the Beclin1-PIK3C3 complex. The decrease of SRSF1 stabilizes the Beclin1 and PIK3C3 complex, activating autophagy, while starvation- and oxidative stresses-induced autophagy can degrade SRSF1 through interacting with LC3-II. This positive feedback mechanism is crucial for inhibiting Gefitinib-resistant cancer cell progression.

Circular RNA circDLC1 inhibits MMP1-mediated liver cancer via interaction with HuR

Hailing Liu et al.

Summary: Our study demonstrates that circDLC1 is downregulated in HCC tissues and is closely associated with a favorable prognosis. Overexpression of circDLC1 inhibits proliferation and motility of hepatoma cells, while silencing of circDLC1 has the opposite effect. Mechanistically, circDLC1 binds to RNA-binding protein HuR, reducing the interaction between HuR and MMP1 mRNA, leading to inhibition of MMP1 expression and ultimately suppressing HCC progression.
