Subthalamic and pallidal deep brain stimulation: are we modulating the same network?

Leon Sobesky et al.

Summary: This study investigated the connectivity profiles of deep brain stimulation electrodes in patients with Parkinson's disease and found a common functional network between subthalamic and pallidal stimulation that is associated with optimal clinical improvements. Furthermore, the study showed that connectivity maps based on different motor symptom subscores can predict treatment outcomes.

Subthalamic and Pallidal Stimulations in Patients with Parkinson's Disease: Common and Dissociable Connections

Chencheng Zhang et al.

Summary: Our study found that STN-DBS and GPi-DBS have similar effects on the motor pathway, but different effects on cortico-cerebellar and somatomotor-visual functional connectivity. This suggests differentiated neural modulation at the two target sites, providing potential mechanistic insights for refining target selection strategies for focal brain stimulation in PD.

Functional analysis of distinct populations of subthalamic nucleus neurons on Parkinson's disease and OCD-like behaviors in mice

Luca Parolari et al.

Summary: The study anatomically characterized molecular markers for specific STN neurons, revealing Pitx2, Gabrr3, Ndnf, and Nos1 expression. Optogenetic studies showed that Gabrr3 neurons play a key role in mediating the beneficial effects of STN modulation, serving as potential cellular targets for PD and OCD drug discovery.
