Recent advances on endogenous/exogenous stimuli-triggered nanoplatforms for enhanced chemodynamic therapy

Qianqian Sun et al.

Summary: This review article presents the latest research progress on enhancing CDT antitumor effects through the amplification strategy of oxidative stress triggered by endogenous/exogenous stimuli, categorizing enhanced CDT nanomaterials into three major groups. Representative examples in each category are dissected and discussed in terms of rational design, mechanism of action, and potential medical applications. The challenges and bottlenecks in current CDT development are summarized, along with preliminary directions for future development in order to maximize the potential power of biology and chemistry coordination.

Recent Progress in Boosted PDT Induced Immunogenic Cell Death for Tumor Immunotherapy

Li Jun et al.

Summary: With the rapid development of materials science, photosensitive materials play an important role in immunogenic cell death (ICD) by inducing reactive oxygen species (ROS) generation under light irradiation. Studies on photodynamic therapy (PDT) induced ICD have shown promising results in cancer immunotherapy, offering various strategies for researchers to explore.

Transformable Nanosensitizer with Tumor Microenvironment-Activated Sonodynamic Process and Calcium Release for Enhanced Cancer Immunotherapy

Xuan Tan et al.

Summary: The study introduces a smart nanosonosensitizer TiO2@CaP which enhances the generation of reactive oxygen species (ROS) under ultrasound activation, inducing mitochondrial dysfunction in the tumor microenvironment and promoting immunogenic cell death, resulting in T-cell infiltration and systemic antitumor immunity. This approach holds promise for developing TME-activatable sonosensitizers with temporospatial control over antitumor responses.

Semiconducting polymer nano-PROTACs for activatable photo-immunometabolic cancer therapy

Chi Zhang et al.

Summary: Proteolysis targeting chimeras (PROTACs) offer an alternative way to modulate protein homeostasis for cancer therapy, but existing PROTACs may have uncontrollable protein degradation and off-target side effects. This study presents a semiconducting polymer nano-PROTAC (SPNpro) with phototherapeutic and activatable protein degradation abilities for photo-immunometabolic cancer therapy. SPNpro can generate singlet oxygen under NIR photoirradiation to eradicate tumor cells and activate protein degradation specifically in response to a cancer biomarker, enhancing tumor immunogenicity.

Zhao Wang et al.

Summary: A biocatalytic Janus nanocomposite, UPFB, was fabricated for ultrasound-driven SDT and 808 nm NIR light-mediated PDT by combining UCNPs and a ferric zirconium porphyrin MOF. UPFB not only promotes ROS generation via GSH depletion and oxygen supply from Fe3+ ions but also achieves CDT performance under acidic conditions. Additionally, UPFB linked with biotin exhibits a good targeting ability to rapidly accumulate in the tumor region, making it suitable for cancer therapeutics.
