Structural and Functional Characterization of Host FHL1 Protein Interaction with Hypervariable Domain of Chikungunya Virus nsP3 Protein

Tetyana Lukash et al.

Summary: Decades of insufficient control have led to unprecedented spread of chikungunya virus (CHIKV), with FHL1 identified as a host factor that interacts with CHIKV nsP3, enhancing viral replication but not being essential. FHL1 and CD2AP have overlapping binding sites in CHIKV nsP3 HVD, with additive effects on viral replication.

The Role of Z-disc Proteins in Myopathy and Cardiomyopathy

Kirsty Wadmore et al.

Summary: The Z-disc is crucial for maintaining the structure and signaling function of the sarcomere in muscle cells. Six key Z-disc proteins, associated with myopathies and cardiomyopathies, play a significant role in force transduction and intracellular signaling. Evaluating the pathogenic variants and minor allele frequency of these proteins in population cohorts can help in re-evaluating their pathogenicity.

The N2A region of titin has a unique structural configuration

Chiara Stronczek et al.

Summary: The N2A segment of titin is a key signaling hub in the sarcomeric I-band, recruiting various signaling factors and processing enzymes. While it has been suggested to play a role in force production through association with actin, the molecular basis for these functions within the titin chain remains poorly understood. N2A has a unique architecture that supports its selective recruitment of binding partners in signaling, but its mechanical role through interaction with F-actin remains to be validated.

Myopathy associated LDB3 mutation causes Z-disc disassembly and protein aggregation through PKCα and TSC2-mTOR downregulation

Pankaj Pathak et al.

Summary: This research demonstrates that the LDB3 protein regulates mechanical stress signaling through interactions with filamin C, HSPA8, and PKC alpha. Mutations in LDB3 lead to early aggregation of filamin C and its chaperones at the muscle Z-disc, ultimately causing protein aggregation myopathy.

Muscle ankyrin repeat protein 1 (MARP1) locks titin to the sarcomeric thin filament and is a passive force regulator

Robbert J. van der Pijl et al.

Summary: This study demonstrates that MARP1 interacts with F-actin and forms a complex with titin-N2A, which leads to the locking of titin to the thin filament, resulting in increased passive force. The mechanism proposed here suggests that in stressed muscle, MARP1 plays a role in protecting the sarcomere from mechanical damage.

Solution NMR Structure of Titin N2A Region Ig Domain I83 and Its Interaction with Metal Ions

Colleen Kelly et al.

Summary: Titin, the largest single chain protein known so far, plays a critical role in passive and active muscle function, with the Ca2+-dependent interaction between its N2A region and the thin filament being key. The structure and dynamics of the I83 domain and its metal binding properties have been characterized, providing insights into its evolutionary optimization for interacting with neighboring domains. A calcium binding site on the N-terminal side of I83 has been identified, which is expected to impact interdomain interaction with the I82 domain.
