Temperature Down-Shift Modifies Expression of UPR-/ERAD-Related Genes and Enhances Production of a Chimeric Fusion Protein in CHO Cells

Summary: Low culture temperature enhances the cell-specific productivity of CHO cells expressing varied r-proteins, potentially through upregulation of UPR-specific transcriptional activators and ER-resident proteins. This results in increased expression of difficult-to-express proteins and improved cell viability, but also leads to decreased cell density and cell cycle arrest. Additionally, low temperature decreases the potential for protein degradation by downregulating genes involved in ERAD.
