Extracellular Matrix Composition Modulates the Responsiveness of Differentiated and Stem Pancreatic Cancer Cells to Lipophilic Derivate of Gemcitabine

Stefania Forciniti et al.

Summary: The study utilized a new potential therapeutic prodrug C18GEM to treat PDAC, showing that PDAC CSCs were more sensitive to C18GEM, especially on collagen I. The mechanism of action of C18GEM was mainly through intracellular uptake via membrane nucleoside transporters and CD36, inducing more cell death and protective autophagy in the cells.

Measuring Both pH and O2 with a Single On-Chip Sensor in Cultures of Human Pluripotent Stem Cell-Derived Cardiomyocytes to Track Induced Changes in Cellular Metabolism

Esther Tanumihardja et al.

Summary: This study introduces a novel use of a ruthenium oxide electrode for in vitro cell metabolism studies, demonstrating its ability to infer different types of metabolism in human pluripotent stem cell-derived cardiomyocytes. The electrode can provide continuous pH readings and measure O-2 concentration over time, allowing for interpretations of extracellular acidification rates and oxygen consumption rates of the cells. The RuOx electrode is presented as a versatile and powerful tool for in vitro cell metabolism studies, particularly in comparative settings.

Investigation of the effect of different chelation solutions on penetration of resin-based and bioceramic sealers with a novel method

Betul Aycan Alim Uysal et al.

Summary: The study found that different chelation solutions can increase the penetration depth of root canal sealers into dentinal tubules. The MA group had the highest penetration depth at the apical third, while the EDTA and HEBP groups had almost equal average depths in the middle and coronal thirds, with significantly higher penetration depth in the coronal thirds of root canals than in other sections. EndoSequence BC Sealer showed higher penetration depth than AH Plus and MTA Fillapex, although the differences were not statistically significant.

Ex vivo Assessment of Mitochondrial Function in Human Peripheral Blood Mononuclear Cells Using XF Analyzer

Alica Schoeller-Mann et al.

Summary: This protocol describes a method to measure key parameters of mitochondrial function in freshly isolated PBMCs using the XF Analyzer technology. By attaching PBMCs to microplates pre-coated with Poly-D-Lysine and injecting stress reagents during OCR and ECAR measurements, various mitochondrial parameters can be calculated. This approach allows the analysis of different influences on human cells, such as pharmaceuticals or environmental factors.
