Bioprinting for the Biologist

Summary: Bioprinting, an emerging technology for building tissues from scratch, has the potential to revolutionize fundamental understanding in biology by exploring entirely new cell configurations. While commonly used in tissue engineering for therapeutic purposes and drug screening, bioprinting also provides opportunities within biology to investigate cellular crosstalk and morphogenesis. This Primer aims to provide an overview of bioprinting for biologists, focusing on extrusion bioprinting as the most widely used technology, and discussing alternative bioprinting technologies and future opportunities in biology.

Recent progress in mechanobiology offers insights into how mechanics regulate cell behaviors and cellular machineries, which can help guide the design of functional engineered tissues. By providing appropriate mechanical cues or interfering with cellular mechanosensing machinery, it may be possible to control cell and tissue structure and function.

Tunable Hybrid Matrices Drive Epithelial Morphogenesis and YAP Translocation

Summary: Morphogenesis is a tightly-regulated developmental process by which tissues acquire critical morphology for their function. Novel hybrid matrices of Matrigel and synthetic oligo(ethylene glycol)-grafted polyisocyanides (PICs) hydrogels are used to create a highly tailorable environment, leading to a broad range of morphologies induced in MDCK cells by controlling stiffness and cell-matrix interactions. The hybrid hydrogels induce a range of morphological changes, including a shift from round hollow cysts to 2D monolayers and the formation of tubular structures in 3D cultures.

This study highlights the complex response of biopolymers and tissue to combined uniaxial and shear strains, providing insights that are likely only obtainable from synthetic model systems. Additionally, it directs further the design of new synthetic biomimetic soft materials that are in high demand as tunable bio-free extracellular matrix materials.

Microrheology reveals simultaneous cell-mediated matrix stiffening and fluidization that underlie breast cancer invasion

Summary: Living tissues are a unique class of hybrid materials where active and thermal forces are closely linked. A new microrheology-based technique was developed to analyze the active and passive fluctuations of the extracellular matrix (ECM) in 3D cell culture models, revealing a dual mechanism of active stiffening and fluidization in breast cancer spheroids. This hybrid dynamics are tightly connected with invasion and suggest a reconciling mechanism for seemingly contradictory requirements in the ECM during invasion by breast cancer cells.

Bioinspired mineralized collagen scaffolds for bone tissue engineering

Summary: This article thoroughly summarizes the state of the art of mineralized collagen scaffolds (MCSs) as tissue-engineered scaffolds for bone repair, including fabrication methods, critical factors for osteogenesis regulation, and opportunities and challenges for the future. The review discusses current fabrication methods for MCSs to improve their biomimetic physical structures, as well as the physical, biological, and chemical cues as critical factors for regulating the osteogenic feature of MCSs. Opportunities and challenges associated with MCSs as bone tissue-engineered scaffolds are also discussed to point out future directions for building the next generation of MCSs with satisfactorily mimetic structures and appropriate biological characters for bone regeneration.
