A Drug Sweeping'' State of the TriABC Triclosan Efflux Pump from Pseudomonas aeruginosa

Summary: The structure of the inner membrane component TriABC and its substrate-free conformation were determined by cryoelectron microscopy, revealing an intermediate step in efflux complex assembly and identifying a tunnel network with constriction that impedes substrate efflux. Functional analyses were used to propose that selective substrate translocation involves conformational gating at the tunnel narrowing.

Tracking the Emergence of Azithromycin Resistance in Multiple Genotypes of Typhoidal Salmonella

Summary: The rising prevalence of antimicrobial resistance in Salmonella enterica serovars Typhi and Paratyphi A, especially against azithromycin, has raised concerns about untreatable infections. Cases of azithromycin-resistant strains with a single point mutation (AcrB-R717Q/L) have been reported in South Asia. Genomic analysis confirmed the spontaneous emergence of this mutation in different genotypes, with a specific genotype predicted to have acquired the mutation in 2010. The development of a low-cost PCR tool for real-time detection of this mutation may aid in early intervention against pan-oral drug resistance.

Cryoelectron Microscopy Structures of AdeB Illuminate Mechanisms of Simultaneous Binding and Exporting of Substrates

Summary: Acinetobacter baumannii, a highly antibiotic-resistant Gram-negative pathogen, utilizes the AdeB multidrug efflux pump to resist a wide range of clinically relevant antimicrobial agents. The study of AdeB pump structures in the presence of ethidium bromide reveals that each AdeB protomer within the trimer can independently recognize and extrude drugs. This sheds light on the drug transport mechanism involving multiple multidrug-binding sites and various transient states of the AdeB membrane protein.

Characteristics and diversity of mutations in regulatory genes of resistance-nodulation-cell division efflux pumps in association with drug-resistant clinical isolates of Acinetobacter baumannii

Summary: The majority of Acinetobacter baumannii isolates in this study were extensively drug-resistant, with efflux pump activity observed in 40% of isolates against multiple antibiotics, mainly tigecycline. Various amino acid substitutions were detected in the products of regulatory genes, with the G186V mutation in AdeS associated with efflux pump overexpression. No insertion sequences were detected. Further studies are needed to understand the exact role of RND efflux pumps in multidrug-resistant clinical isolates of A. baumannii.
