Liposomes and Extracellular Vesicles as Drug Delivery Systems: A Comparison of Composition, Pharmacokinetics, and Functionalization

Luke van der Koog et al.

Summary: Liposomes and extracellular vesicles (EVs) are two important nanoparticle drug delivery systems, composed of lipid bilayers and biologically secreted nanoparticles, each with its own advantages. Research has evaluated the differences and similarities in their composition, as well as critical mediators affecting pharmacokinetics; exploring different strategies to tweak their pharmacokinetics further demonstrates the potential advantages of these systems in drug delivery.

Challenges and pitfalls in the development of liposomal delivery systems for cancer therapy

Seyedeh Alia Moosavian et al.

Summary: This review highlights the challenges in translating liposomal delivery systems from preclinical studies to clinical application in cancer therapy. The gap between experimental results and practical use of liposomes is a major concern that needs to be addressed in order to improve the efficacy of this drug delivery system. Various potential approaches to overcome the barriers in clinical application of liposomal formulations are also discussed.

Copper metabolism as a unique vulnerability in cancer

Vinit C. Shanbhag et al.

Summary: Over the past 25 years, significant progress has been made in identifying and characterizing proteins that regulate the uptake, intracellular trafficking and export of copper. While copper is essential for growth and development in humans and other mammals, it is also a limiting nutrient for cancer cell growth and proliferation. Oral copper chelators have been shown to have anti-neoplastic and anti-metastatic effects in animals and humans.

Seesaw conformations of Npl4 in the human p97 complex and the inhibitory mechanism of a disulfiram derivative

Man Pan et al.

Summary: The human AAA+protein p97 plays a crucial role in cellular protein homeostasis. The study reveals insights into the interaction between p97 and its co-factor Npl4, showing three distinct conformational states of Npl4 in complex with human p97, suggesting the essential role of seesaw motion in the unfolding activity of the p97 complex.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Disulfiram-loaded functionalized magnetic nanoparticles combined with copper and sodium nitroprusside in breast cancer cells

Kubra Solak et al.

Summary: Disulfiram, a reactive species generator, can induce cancer cell death in breast cancer but it degrades easily in blood. This study prepared a drug carrier system based on magnetic mesoporous silica nanoparticles to support selective accumulation of DSF in tumor cells. The system showed high drug-loading capacity and achieved cancer-selective accumulation through modifications with folic acid linked polyethyleneimine. The cytotoxicity of the system on breast cancer cells was higher than on non-tumorigenic cells due to enhanced uptake by cancer cells.

EMT and Cancer Cell Stemness Associated With Chemotherapeutic Resistance in Esophageal Cancer

Xiaojie Liu et al.

Summary: Our study revealed the presence of cancer cell stemness characteristics in drug-resistant esophageal cancer cells, expressing stem cell biomarkers and undergoing epithelial-mesenchymal transition (EMT). The increased expression of EMT biomarkers and stemness-related proteins in esophageal cancer cells after prolonged exposure to chemotherapeutic drugs suggests that targeting EMT and stemness simultaneously could be an effective strategy for treating esophageal cancer drug resistance.
