Auranofin Resistance in Toxoplasma gondii Decreases the Accumulation of Reactive Oxygen Species but Does Not Target Parasite Thioredoxin Reductase

Summary: Auranofin, initially used for rheumatoid arthritis, has shown promise as an anti-parasitic drug against Toxoplasma gondii. Resistance to auranofin in T. gondii was not associated with mutations in its predicted target, thioredoxin reductase, but instead with mutations in genes encoding redox-relevant proteins. This resistance was found to result in lower accumulation of reactive oxygen species in the resistant clones compared to the wild type strains.
