Modulation of the tumor microenvironment by natural agents: implications for cancer prevention and therapy

Haseeb Zubair et al.

Summary: The development of cancer involves the coevolution of tumor cells with their surrounding environment, and the interactions between tumor cells and stromal cells play a crucial role in tumor growth, metastasis, and therapy resistance. Natural compounds have significant potential in targeting the tumor microenvironment.

Immune suppression in gliomas

Matthew M. Grabowski et al.

Summary: Gliomas evade immune detection and destruction through mechanisms such as dysfunctional T cells and NK cells, expansion of regulatory T cells and myeloid-derived suppressor cells, M2 phenotypic transformation in macrophages/microglia, upregulation of immunosuppressive factors, tumor microenvironment hypoxia, and iatrogenic effects of immunosuppressive treatments. Future research should focus on addressing these immunosuppressive mechanisms to develop treatment options with improved survival outcomes.

Exploring the multifocal role of phytochemicals as immunomodulators

Tapan Behl et al.

Summary: The well-functioning immune system of the host body is crucial for maintaining normal physiological and immunological functions, as well as the internal environment. Balancing immunity enhances defense mechanisms against infections, diseases, and unwanted pathogens to prevent hypersensitivity reactions and immune-related diseases. Recent interest in the immune system as a significant target for toxicity has increased due to exposure to chemicals, drugs, and environmental pollutants. Various factors such as sex, age, stress, malnutrition, alcohol, genetic variability, lifestyles, environmental pollutants, and chemotherapy exposure can alter immune responses. Immunomodulation involves modifying immune responses through induction, amplification, attenuation, or inhibition. Phytochemicals, including glycosides, alkaloids, phenolic acids, flavonoids, saponins, tannins, sterols, and sterolins, play a key role in modulating favorable immune responses.

Triptolide suppresses oral cancer cell PD-L1 expression in the interferon-γ-modulated microenvironment in vitro, in vivo, and in clinical patients

Chin-Shan Kuo et al.

Summary: The study found significant overexpression of PD-L1 in oral squamous cell carcinoma tissues, and that IFN-gamma can drive PD-L1 expression in OSCC cells. Triptolide (TPL) can suppress oral cancer growth and downregulate PD-L1 expression in oral cancer cells in vitro.

Icaritin inhibits PD-L1 expression by Targeting Protein IκB Kinase α

Dongliang Mo et al.

Summary: Icaritin, a prenylflavonoid derivative from the Epimedium genus, has been identified to target IKK-alpha and inhibit the NF-kappa B signaling pathway in treating advanced hepatocellular carcinoma. It downregulates PD-L1 expression and shows improved efficacy when combined with checkpoint antibodies in animal models.

Luteolin suppresses androgen receptor-positive triple-negative breast cancer cell proliferation and metastasis by epigenetic regulation of MMP9 expression via the AKT/mTOR signaling pathway

Han-Tsang Wu et al.

Summary: The study showed that luteolin effectively inhibited the proliferation and metastasis of androgen receptor-positive TNBC, reversing the epithelial-mesenchymal transition and downregulating MMP9 expression. The combination of luteolin and AKT/mTOR inhibitors synergistically suppressed the proliferation and metastasis of androgen receptor-positive TNBC cells.
