Uncertainties in estimating SARS-CoV-2 prevalence by wastewater-based epidemiology

Xuan Li et al.

Summary: Wastewater-based epidemiology (WBE) is a promising approach for estimating COVID-19 prevalence by detecting SARS-CoV-2 RNA in wastewater, but methodological challenges could impact accuracy. Dividing WBE into five steps, this study quantified uncertainties at each step and found that using high-frequency flow-proportional sampling and actual water usage data reduced overall uncertainties to 20-40%, with analytical uncertainty being the dominant factor. Using surrogate viruses and improving analytical approaches could enhance reliability of WBE for monitoring COVID-19 prevalence.

Comparison of approaches to quantify SARS-CoV-2 in wastewater using RT-qPCR: Results and implications from a collaborative inter-laboratory study in Canada

Alex H. S. Chik et al.

Summary: Detection of SARS-CoV-2 RNA in wastewater can inform public health decisions during the COVID-19 pandemic, but the analysis methods are not yet standardized globally. Research in Canada showed that different methods yielded comparable results with low variability, suggesting the need for a consistent method for evaluating wastewater SARS-CoV-2 temporal trends.

Estimating epidemiologic dynamics from cross-sectional viral load distributions

James A. Hay et al.

Summary: Population distribution of viral loads changes during an epidemic, and Ct values from random samples can improve estimates of an epidemic's trajectory. Combining data from multiple samples enhances the precision and robustness of estimation. These methods can be applied to real-time estimates of epidemic trajectories for outbreak management and response.

Polio eradication at the crossroads

Konstantin Chumakov et al.

Summary: The initiative to eradicate polio needs a reassessment of its strategy and proposes a new strategic goal of eradicating the disease rather than the virus. This can be achieved by implementing global immunisation programmes with improved vaccines to create and maintain high rates of population immunity indefinitely.

Wastewater-Based Epidemiology for Managing the COVID-19 Pandemic

Summary: Testing wastewater for SARS-CoV-2 provides information on disease prevalence, infection trends, hot spots, and helps decision makers. Studies also examine virus survivability in different water matrices, the impact of wastewater treatment processes, and the potential of wastewater-based epidemiology.
