Cell-specific characterization of the placental methylome

Victor Yuan et al.

Summary: This research conducted a comprehensive DNA methylation analysis of human placental cell types, revealing unique methylation profiles in trophoblasts and Hofbauer cells that impact placental development and pregnancy-related diseases. The results provide valuable methylation reference data for future placental studies.

Inherent mosaicism and extensive mutation of human placentas

Tim H. H. Coorens et al.

Summary: Placentas can exhibit chromosomal abnormalities different from the fetus, possibly due to genetic segregation. Research shows that placental cells display genetic isolation and extensive mutagenesis, suggesting that mosaicism is a typical feature of placental development.

Human naive epiblast cells possess unrestricted lineage potential

Ge Guo et al.

Summary: Classic embryological experiments have shown that early mouse embryos develop through sequential lineage bifurcations. However, human naive embryonic stem cells exhibit the ability to produce blastocyst trophectoderm, unlike their mouse counterparts. This suggests a difference in developmental specification and lineage restriction between human and mouse embryos.

Capturing human trophoblast development with naive pluripotent stem cells in vitro

Shingo Io et al.

Summary: The study demonstrates that naive human pluripotent stem cells can recapitulate trophoblast development, establishing an authentic paradigm for human trophoblast development. These findings provide valuable insights into the molecular mechanisms underlying trophoblast development and related diseases.

Modeling preeclampsia using human induced pluripotent stem cells

Mariko Horii et al.

Summary: Preeclampsia is a pregnancy-specific hypertensive disorder affecting up to 10% of pregnancies worldwide, primarily caused by abnormal development and function of placental cells. Research on induced pluripotent stem cells derived from placentas with or without preeclampsia revealed defects in syncytialization and hypoxia response, indicating the importance of maternal microenvironment in normal placentation and potential pathways for diagnosis or therapy. Marked DNA methylation changes were minimal, suggesting other regulatory mechanisms mediate these alterations.

Two distinct trophectoderm lineage stem cells from human pluripotent stem cells

Adam Mischler et al.

Summary: The trophectoderm layer of the blastocyst-stage embryo is the precursor for all trophoblast cells in the placenta. Human trophoblast stem (TS) cells have emerged as an attractive tool for studies on early trophoblast development. The derivation of TS cells from pluripotent stem cells will significantly enable construction of in vitro models for normal and pathological placental development.
