DNA Double-Strand Break Accumulation in Alzheimer's Disease: Evidence from Experimental Models and Postmortem Human Brains

Summary: Alzheimer's disease (AD) is characterized by progressive neurodegeneration with accumulation of DNA double-strand breaks (DSBs) playing an important role in its pathogenesis. Studies have shown increased DNA DSB accumulation and reduced repair function in the hippocampus of AD brains compared to non-AD brains. Additionally, altered levels of DNA repair proteins were observed in AD mouse models and cellular models, highlighting the potential influence of DNA DSB and repair proteins in the pathway toward neural damage and memory loss in AD.

Summary: Through single-cell RNA sequencing, a subpopulation of microglia named sMG2 highly expressed necroptosis-related genes Rip3 and Mlkl, playing a role in retinal neovascularization. The hypoxia-induced microglial activation committed to necroptosis through the RIP1/RIP3-mediated pathway, which significantly decreased retinal angiogenesis. Targeting the microglia necroptosis axis is shown to be an effective antiangiogenesis therapy for retinal neovascular diseases, especially when combined with VEGF blockade.
