A guide to: generation and design of nanobodies

Summary: Nanobodies, a trademark of Ablynx, are single antigen-binding domains of heavy chain-only antibodies in Camelidae produced recombinantly for research and medical applications. Various methods are used for their generation, producing cost-effective Nbs with high affinity for their target antigens. Additionally, engineering technologies have been explored to broaden the application range of Nbs for potential advantages over other affinity reagents.

Peste des Petits Ruminants Virus Infection at the Wildlife-Livestock Interface in the Greater Serengeti Ecosystem, 2015-2019

Summary: Evidence of PPRV infection was found in wild artiodactyl species in the Greater Serengeti and Amboseli ecosystems of Kenya and Tanzania. The relatively low seroprevalence suggests that sustained transmission of PPRV between wild animals is unlikely, with transmission possibly occurring through spillover from infected small ruminants. Further study of other major wild artiodactyl species in this ecosystem is needed.

Review of Peste des Petits Ruminants Occurrence and Spread in Tanzania

The disease has not been observed in wildlife, cattle, or camels, but they show exposure to PPRV through serological evidence. Challenges for PPR control in Tanzania include small ruminant movements and limited veterinary services, highlighting the need for a comprehensive eradication program.

This study presents a simple directed evolution method for producing nanobodies with high affinities and neutralization activities, which can effectively neutralize both SARS-CoV-2 pseudovirus and live virus.

Antiviral Potential of Ivermectin against Peste des Petits Ruminants Virus (PPRV)

Summary: The study showed that 2.5 mu M concentration of ivermectin is non-toxic to Vero cells and significantly reduces the titre of Peste des Petits Ruminants Virus (PPRV) in vitro. Additionally, ivermectin exposure after viral attachment and entry steps is more effective in reducing virus titre, indicating a possible mechanism of action for the drug.
