Whole-genome sequencing to detect mutations associated with resistance to insecticides and Bt proteins inSpodoptera frugiperda

Fang Guan et al.

Summary: The study identified three amino acid substitutions in the ace-1 gene of S. frugiperda associated with organophosphate resistance, with the Brazilian population having all three mutations and the 227A allele being the most common. Populations from China, Malawi, and Uganda carried two of the three ace-1 mutations, with the 290V allele as the dominant allele.

Toxicological and molecular profiling of insecticide resistance in a Brazilian strain of fall armyworm resistant to Bt Cry1 proteins

Debora Boaventura et al.

Summary: The study found that the Bt-resistant Brazilian strain Sf_Des has medium resistance to deltamethrin and chlorpyrifos, high cross-resistance to Cry1 toxins but high susceptibility to Vip3A. In comparison to the susceptible strain Sf_Bra, Sf_Des exhibited up to 19-fold increased cytochrome P450 activity and upregulation of multiple P450 transcripts. Gene Ontology analysis categorized most differentially expressed genes into oxidation-reduction and metabolic processes.

Mechanisms of Resistance to Insecticidal Proteins from Bacillus thuringiensis

Juan Luis Jurat-Fuentes et al.

Summary: Insecticidal proteins from Bt bacterium are widely used as successful alternatives to synthetic pesticides, but the evolution of resistance in target pests remains a major threat. High-level resistance to Bt sprays is limited in the field, but several lepidopteran and coleopteran species have developed practical resistance to Bt proteins produced in transgenic plants.

Two ABC transporters are differentially involved in the toxicity of two Bacillus thuringiensis Cry1 toxins to the invasive crop-pest Spodoptera frugiperda (J. E. Smith)

Minghui Jin et al.

Summary: In this study, two potential functional Bt receptors, SfABCC2 and SfABCC3, were created in the fall armyworm using CRISPR/Cas9 genome editing system, showing that SfABCC2 plays a major role in the toxicity of Cry1Fa and Cry1Ab toxins. Additionally, the knockout of SfABCC2 or SfABCC3 increased susceptibility to abamectin and spinosad insecticides.

Transcriptional regulation of xenobiotic detoxification genes in insects-An overview

Dries Amezian et al.

Summary: Arthropods have evolved various strategies, including the induction of detoxification enzymes, to defend themselves against a wide array of chemicals in their environment. Studies using advanced genetic tools in insects have substantially contributed to our understanding of the genomic basis of their xenobiotic response. Signaling pathways such as nuclear receptors and transcription factors play crucial roles in the upregulation of detoxification enzymes and can impact insects' adaptive processes.

Changes in both trans- and cis-regulatory elements mediate insecticide resistance in a lepidopteron pest, Spodoptera exigua

Bo Hu et al.

Summary: The overexpression of P450 gene CYP321A8 in the insect Spodoptera exigua, resistant to multiple insecticides, was identified as a major factor contributing to resistance. Two independent mechanisms, constitutive upregulation of transcription factors and mutations in the promoter region allowing for binding of an orphan nuclear receptor, were found to be involved in the overexpression of CYP321A8 and resistance in S. exigua.

Summary: The genus Spodoptera consists of 31 species with varied host plant breadths, with some species being major economic pests. Comparative genomics studies have highlighted major adaptive changes in genetic architecture among different Spodoptera species. Recent population genomics studies provide insights into population structures, migration patterns, and invasion processes of two species.
