In vitro model for predicting the access and distribution of drugs in the brain using hCMEC/D3 cells

Summary: This study aimed to improve a previously proposed in vitro cell-based model by using a more physiological cell line and established four linear IVIVCs to predict the pharmacokinetic parameters related to drug access and distribution in the CNS. The results showed that the best IVIVCs were those correlating in vitro and in vivo parameters related to the binding rate of a substance to brain tissue, highlighting the importance of using brain homogenate in in vitro brain permeability assays.
