Fuchs endothelial corneal dystrophy: The vicious cycle of Fuchs pathogenesis

Summary: Fuchs endothelial corneal dystrophy (FECD) is the most common primary corneal endothelial dystrophy characterized by the decline of corneal endothelial cells and the formation of extracellular matrix excrescences, leading to corneal edema and loss of vision. It is a complex and heterogeneous genetic disease with a higher incidence in women, caused by a combination of genetic and environmental factors.

Knockout of c-Cbl slows EGFR endocytic trafficking and enhances EGFR signaling despite incompletely blocking receptor ubiquitylation

Summary: Preventing the downregulation of EGFR is proposed as an alternative mechanism for enhancing signaling and promoting corneal restoration. Knockout of c-Cbl leads to prolonged EGFR phosphorylation and sustained signaling, resulting in enhanced downstream signaling with altered receptor ubiquitylation and endocytic trafficking. MAPK phosphorylation remains sustained for a longer period in c-Cbl knockout cells.
