eIF2B conformation and assembly state regulate the integrated stress response

Summary: The integrated stress response (ISR) is activated by phosphorylation of translation initiation factor eIF2 in response to various stress conditions. Phosphorylated eIF2 inhibits eIF2B, leading to ISR induction. ISRIB can inhibit ISR even in fully assembled eIF2B decamers, suggesting allosteric communication between distant binding sites.
